Safety: Work only under teacher supervision using approved low-risk teaching strains and your school's microbiology procedures. Keep plates closed after incubation unless instructed otherwise, disinfect the work area, and dispose of cultures as directed by your teacher.
1
Preparing Agar Plates
Objective: Create sterile nutrient agar plates for bacterial cultivation.
Procedure reference
- Weigh 2.8 g agar powder and add to 100 mL distilled water.
- Sterilize by autoclaving at 121°C for 15 minutes.
- Carefully pour into sterile Petri dishes using teacher-approved aseptic technique.
- Allow plates to solidify for about 30 minutes.
- Store plates inverted as directed by your teacher.

2
Streak Plate Method
Objective: Isolate individual bacterial colonies using aseptic technique.
Procedure reference
- Sterilize the inoculating loop as directed.
- Collect bacteria from the approved broth culture.
- Streak Zone 1.
- Re-sterilize/cool the loop and streak Zone 2 from Zone 1.
- Repeat for Zone 3.
- Incubate at 25°C for 24–48 hours, or as directed by your teacher.

Gram Staining
Objective: Differentiate bacteria based on cell wall structure.
Procedure reference
- Prepare and heat-fix a bacterial smear.
- Apply crystal violet for 60 s, then rinse.
- Apply Gram's iodine for 60 s, then rinse.
- Decolorize briefly with alcohol, then rinse immediately.
- Apply safranin for 60 s, rinse and blot dry.
- Observe using oil immersion at approximately 1000× total magnification.
